Penetration of active substances through the skin. Factors affecting this phenomenon and methods for its modification

ABSTRACT
The skin is the largest organ of the human body. Its condition depends on many internal and external factors. The organ performs many important functions, including isolation of the body’s interior from the external environment, participation in thermoregulation process of the body and the perception of stimuli. The skin is an important transport route for some active substances. The aim of the article was to present the mechanism of penetration of active substances through the epidermal barrier and factors deteminating this process. There are many methods to increase the penetration of molecules across the epidermal barrier. Some of them are related to the active substance (increasing lipophilicity by modifying the molecule, using carriers such as nanocapsules or liposomes). The other methods focus on changing the properties or thickness of the stratum corneum. In the first case, the socalled permeation promoters, current or ultrasound are being used. In the second, a specific part of the stratum corneum can be removed using mechanical (microdermabrasion, cavitation peeling) and chemical peels (so-called chemexfoliation). At the stage of designing a cosmetic, it is worth carrying out in vitro and in vivo tests to assess the penetrating ability of the active substance molecules, as well as the effectiveness of its action.

STRESZCZENIE
Skóra to największy organ ludzkiego ciała, którego stan zależy od wielu czynników wewnątrz- i zewnątrzpochodnych. Organ ten pełni wiele istotnych funkcji, m.in. izoluje wnętrze organizmu od środowiska zewnętrznego, bierze udział w termoregulacji organizmu oraz percepcji bodźców. Skóra stanowi istotną drogę transportu niektórych substancji aktywnych. Celem artykułu było przedstawienie mechanizmu przenikania substancji aktywnych przez barierę naskórkową oraz czynników determinujących ten proces. Istnieje wiele metod mających na celu zwiększenie przenikania cząsteczek przez barierę naskórkową. Część z nich jest związana z substancjami aktywnymi – zwiększenie lipofilowości poprzez modyfikację cząsteczki, wykorzystanie nośników typu nanokapsułki czy liposomy. Pozostałe metody skupiają się na zmianie właściwości lub grubości warstwy rogowej. W pierwszym przypadku wykorzystuje się tzw. promotory przenikania, prąd lub ultradźwięki. W drugim z kolei usuwa się określoną część warstwy rogowej naskórka wykorzystując w tym celu peelingi mechaniczne (mikrodermabrazja, peeling kawitacyjny) oraz chemiczne (tzw. chemeksfoliacja). Na etapie projektowania kosmetyku warto przeprowadzić testy in vitro oraz in vivo, które mają na celu oszacowanie zdolności penetracyjnych cząsteczek substancji aktywnej, a także skuteczności jej działania.
